Functional classes of bronchial mucosa genes that are differentially expressed in asthma - GSE15823

Purpose

Asthma pathogenesis and susceptibility involves a complex interplay between genetic and environmental factors.We report the results of expression microarray studies using tissue obtained from bronchial biopsies of healthy controls and of subjects with allergic asthma, both before and following inhaled corticotherapy.

Hypothesis

Transcriptional profile in bronchial biopsies between healthy and asthmatic individual differs and implicate novel genes and genes previously known to play a role in asthma.

Experimental Design
Four healthy subjects (2 men and 2 women) without a history of allergy or asthma and four asthmatics (all women) meeting the criteria of the American Thoracic Society for the diagnosis of asthma were recruited to the study. Microarray expression studies were performed using Affymetrix HG-U95Av2 GeneChips.

Methods

Biopsies were obtained from the sub-segmental and segmental bronchial carinae of the lower (3–5 biopsies) and upper (1–2 biopsies) lobes, as well as the lobar carinae of the middle (2 biopsies) and upper (1–2 biopsies) lobes to provide adequate samples of the large airways (3rd to 5th bronchial generation). When bronchoscopy was repeated after ICS treatment, biopsies were taken from the left lung at similar bronchial levels. The samples were mechanically homogenized (Rotor/Stator homogeniser, PowerGen) and total RNA was extracted using the RNEasy kit (Qiagen) according to the manufacturer's instructions.
